IPR001577

Description

IPR001577 is a Peptidase M8, leishmanolysin.

This group of metallopeptidases belong to the MEROPS peptidase family M8 (leishmanolysin family, clan MA(M)). The protein fold of the peptidase domain for members of this family resembles that of thermolysin, the type example for clan MA.

Leishmanolysin ([ec:3.4.24.36]) is an enzyme found in eukaryotes including Leishmania and related parasitic protozoa [[cite:PUB00003579]]. The endopeptidase is the most abundant protein on the cell surface during the promastigote stage of the parasite, and is attached to the membrane by a glycosylphosphatidylinositol anchor [[cite:PUB00003579]]. In the amastigote form, the parasite lives in lysosomes of host macrophages, producing a form of the protease that has an acidic pH optimum [[cite:PUB00003579]]. This differs from most other metalloproteases and may be an adaptation to the environment in which the organism survives [[cite:PUB00003579]].

Tris entry also includes proteins from humans and other chordates, including Leishmanolysin-like peptidase or invadolysin [[cite:PUB00090184]] and Ciliated left-right organizer metallopeptidase, CIROP). CIROP plays a role in left-right patterning process [[cite:PUB00101066]].

Associated GO terms

GO predictions are based solely on the InterPro-to-GO mappings published by EMBL-EBI, which are in turn based on the mapping of predicted domains to the InterPro dataset. The InterPro-to-GO mapping was last updated on , while the GO metadata was last updated on .

GO term Namespace Name Definition Relationships
Molecular function Metalloendopeptidase activity Catalysis of the hydrolysis of internal, alpha-peptide bonds in a polypeptide chain by a mechanism in which water acts as a nucleophile, one or two metal ions hold the water molecule in place, and charged amino acid side chains are ligands for the metal ions.
Biological process Proteolysis The hydrolysis of proteins into smaller polypeptides and/or amino acids by cleavage of their peptide bonds.
Biological process Cell adhesion The attachment of a cell, either to another cell or to an underlying substrate such as the extracellular matrix, via cell adhesion molecules.
Cellular component Membrane A lipid bilayer along with all the proteins and protein complexes embedded in it an attached to it.
