IPR004393 is a Nicotinate-nucleotide pyrophosphorylase.

Nicotinate-nucleotide pyrophosphorylase ([ec:2.4.2.19]), also known as quinolinate phosphoribosyltransferase (decarboxylating), catalyses the conversion of nicotinate D-ribonucleotide, pyrophosphate and carbon dioxide into pyridine-2,3-dicarboxylate and 5-phospho-alpha-D-ribose 1-diphosphate. This enzyme is a type II phosphoribosyltransferase which provides thede novosource of nicotinate mononucleotide (NAMN) for NAD biosynthesis in both prokaryotes and eukaryotes [[cite:PUB00017199], [cite:PUB00017200]].

Structural studies have shown that the active form of this enzyme is a homodimer with an unsual fold [[cite:PUB00005289], [cite:PUB00017201], [cite:PUB00017204]]. The N-terminal forms a mixed α/β domain, while the C-terminal region forms a multi-stranded, open α/β-barrel. The active site is found at the C-terminal ends of the β-strands of the α/β-barrel, and is bordered by the N-terminal domain of the second subunit of the dimer. It contains several conserved charged residues thought to be important determinants of substrate binding and catalysis.

Molecular function | Nicotinate-nucleotide diphosphorylase (carboxylating) activity | Catalysis of the reaction: CO(2) + diphosphate + nicotinate D-ribonucleotide = 5-phospho-alpha-D-ribose 1-diphosphate + 2 H(+) + quinolinate. | ||
Biological process | NAD biosynthetic process | The chemical reactions and pathways resulting in the formation of nicotinamide adenine dinucleotide, a coenzyme present in most living cells and derived from the B vitamin nicotinic acid; biosynthesis may be of either the oxidized form, NAD, or the reduced form, NADH. |
