IPR019160

Description

IPR019160 is a Exocyst complex component Sec3, coiled-coil.

This entry represents the C-terminal domain of Sec3 (also known as ExoC1), a component of the exocyst complex (composed of Exoc1, Exoc2, Exoc3, Exoc4, Exoc5, Exoc6, Exoc7 and Exoc8) which mediates the tethering of post-Golgi secretory vesicles to the plasma membrane and promotes the assembly of the SNARE complex for membrane fusion. The exocyst is also involved in other cell processes such as cell polarisation, primary ciliogenesis, cytokinesis, and tumorigenesis and metastasis [[cite:PUB00045127]]. This complex has an elongated shape consisting of packed long rods, a structure that is shared among the Complex Associated with Tethering Containing Helical Rods (CATCHRs) proteins from related complexes such as Conserved Oligomeric Golgi complex (COG) and Golgi-Associated Retrograde Protein complex (GARP) [[cite:PUB00100040], [cite:PUB00100048], [cite:PUB00100047]]. Subunits of these complexes, apart of helical bundles, they usually have a coiled-coil (CC) region at the N-terminal. Sec3 is described as a membrane-anchoring component which serves as a spatial landmark in the plasma membrane for incoming secretory vesicles. Sec3 binds to the C-terminal cytoplasmic domain of GLYT1 (glycine transporter protein 1). Sec3 is recruited to the sites of polarised membrane growth through its interaction with Rho1p, a small GTP-binding protein [[cite:PUB00045127]].

Associated GO terms

GO predictions are based solely on the InterPro-to-GO mappings published by EMBL-EBI, which are in turn based on the mapping of predicted domains to the InterPro dataset. The InterPro-to-GO mapping was last updated on , while the GO metadata was last updated on .

GO term Namespace Name Definition Relationships
Cellular component Exocyst A protein complex peripherally associated with the plasma membrane that determines where vesicles dock and fuse. At least eight complex components are conserved between yeast and mammals.
Biological process Exocytosis A process of secretion by a cell that results in the release of intracellular molecules (e.g. hormones, matrix proteins) contained within a membrane-bounded vesicle. Exocytosis can occur either by full fusion, when the vesicle collapses into the plasma membrane, or by a kiss-and-run mechanism that involves the formation of a transient contact, a pore, between a granule (for exemple of chromaffin cells) and the plasma membrane. The latter process most of the time leads to only partial secretion of the granule content. Exocytosis begins with steps that prepare vesicles for fusion with the membrane (tethering and docking) and ends when molecules are secreted from the cell.
