IPR035435

Description

IPR035435 is a Diphthamide synthesis DHP1/DPH2, eukaryotes and archaea.

This entry includes diphthamide synthesis DPH1 from eukaryotes and archaea and DPH2 from archaea.

Archaeal and eukaryotic translation elongation factor 2 (EF-2) contain a unique posttranslationally modified histidine residue called diphthamide, which become the target for ADP-ribosylation by diphtheria toxin [[cite:PUB00017070]]. DPH1 catalyzes the first step of diphthamide biosynthesis and is required for the modification of EF-2 [[cite:PUB00017070]]. The human DPH1 is also known as OVCA1 (for ovarian cancer gene 1), a tumour suppressor gene that plays a crucial role in the regulation of cell proliferation, embryonic development, and tumorigenesis [[cite:PUB00085370]].

Associated GO terms

GO predictions are based solely on the InterPro-to-GO mappings published by EMBL-EBI, which are in turn based on the mapping of predicted domains to the InterPro dataset. The InterPro-to-GO mapping was last updated on , while the GO metadata was last updated on .

GO term Namespace Name Definition Relationships
Biological process Peptidyl-diphthamide biosynthetic process from peptidyl-histidine The modification of peptidyl-histidine to 2'-(3-carboxamido-3-(trimethylammonio)propyl)-L-histidine, known as diphthamide, found in translation elongation factor EF-2. The process occurs in eukaryotes and archaea but not eubacteria.
