IPR036605

Description

IPR036605 is a Mago nashi superfamily.

This superfamily was originally identified in Drosophila and called mago nashi, it is a strict maternal effect, grandchildless-like, gene [[cite:PUB00007284]]. The protein is an integral member of the exon junction complex (EJC). The EJC is a multiprotein complex that is deposited on spliced mRNAs after intron removal at a conserved position upstream of the exon-exon junction, and transported to the cytoplasm where it has been shown to influence translation, surveillance, and localization of the spliced mRNA. It consists of four core proteins (eIF4AIII, Barentsz [Btz], Mago, and Y14), mRNA, and ATP and is supposed to be a binding platform for more peripherally and transiently associated factors along mRNA travel. Mago and Y14 form a stable heterodimer that stabilizes the complex by inhibiting eIF4AIII's ATPase activity. Mago-Y14 heterodimer has been shown to interact with the cytoplasmic protein PYM, an EJC disassembly factor, and specifically binds to the karyopherin nuclear receptor importin 13 [[cite:PUB00079498], [cite:PUB00079499], [cite:PUB00079500], [cite:PUB00079501], [cite:PUB00029696], [cite:PUB00079502], [cite:PUB00079503], [cite:PUB00007284], [cite:PUB00007286], [cite:PUB00007285], [cite:PUB00019831], [cite:PUB00079504], [cite:PUB00021760], [cite:PUB00030702], [cite:PUB00079505], [cite:PUB00079506], [cite:PUB00079507]].

The human homologue has been shown to interact with an RNA binding protein, ribonucleoprotein rbm8 ([swissprot:Q9Y5S9]) [[cite:PUB00007285]]. An RNAi knockout of the Caenorhabditis elegans homologue causes masculinization of the germ line (Mog phenotype) hermaphrodites, suggesting it is involved in hermaphrodite germ-line sex determination [[cite:PUB00007286]] but the protein is also found in hermaphrodites and other organisms without a sexual differentiation.

Structurally, Mago nashi has a β(4)-α-β(2)-α fold arranged into two layers (α/β) with an antiparallel β-sheet.

GO term Namespace Name Definition Relationships
Cellular component Nucleus A membrane-bounded organelle of eukaryotic cells in which chromosomes are housed and replicated. In most cells, the nucleus contains all of the cell's chromosomes except the organellar chromosomes, and is the site of RNA synthesis and processing. In some species, or in specialized cell types, RNA metabolism or DNA replication may be absent.
