IPR008047

Description

IPR008047 is a Mini-chromosome maintenance complex protein 4.

<p>The MCM2-7 complex consists of six closely related proteins that are highly conserved throughout the eukaryotic kingdom. In eukaryotes, Mcm3 is a component of the MCM2-7 complex (MCM complex), which consists of six sequence-related AAA + type ATPases/helicases that form a hetero-hexameric ring [[cite:PUB00063047]]. MCM2-7 complex is part of the pre-replication complex (pre-RC). In G1 phase, inactive MCM2-7 complex is loaded onto origins of DNA replication [[cite:PUB00063043], [cite:PUB00063044], [cite:PUB00063046]]. During G1-S phase, MCM2-7 complex is activated to unwind the double stranded DNA and plays an important role in DNA replication forks elongation [[cite:PUB00063046]].</p> <p>The components of the MCM2-7 complex are:</p> <ul> <li>DNA replication licensing factor MCM2, [interpro:IPR008045]</li> <li>DNA replication licensing factor MCM3, [interpro:IPR008046]</li> <li>DNA replication licensing factor MCM4, [interpro:IPR008047]</li> <li>DNA replication licensing factor MCM5, [interpro:IPR008048]</li> <li>DNA replication licensing factor MCM6, [interpro:IPR008049]</li> <li>DNA replication licensing factor MCM7, [interpro:IPR008050]</li> </ul> <p>Mcm4 is thought to play a pivotal role in ensuring DNA replication occurs only once per cell cycle. Phosphorylation of Mcm4 dramatically reduces its affinity for chromatin - it has been proposed that this cell cycle-dependent phosphorylation is the mechanism that inactivates the MCM complex from late S phase through mitosis, thus preventing illegitimate DNA replication during that period of the cell cycle [[cite:PUB00010306]].</p>

Associated GO terms

GO predictions are based solely on the InterPro-to-GO mappings published by EMBL-EBI, which are in turn based on the mapping of predicted domains to the InterPro dataset. The InterPro-to-GO mapping was last updated on , while the GO metadata was last updated on .

GO term Namespace Name Definition Relationships
Molecular function DNA binding Any molecular function by which a gene product interacts selectively and non-covalently with DNA (deoxyribonucleic acid).
Molecular function DNA helicase activity Catalysis of the reaction: NTP + H2O = NDP + phosphate, to drive the unwinding of a DNA helix.
Molecular function ATP binding Interacting selectively and non-covalently with ATP, adenosine 5'-triphosphate, a universally important coenzyme and enzyme regulator.
Biological process DNA replication initiation The process in which DNA-dependent DNA replication is started; this begins with the ATP dependent loading of an initiator complex onto the DNA, this is followed by DNA melting and helicase activity. In bacteria, the gene products that enable the helicase activity are loaded after the initial melting and in archaea and eukaryotes, the gene products that enable the helicase activity are inactive when they are loaded and subsequently activate.
Cellular component MCM complex A hexameric protein complex required for the initiation and regulation of DNA replication.
