This group also includes the tRNA nucleotidyltransferase that adds the CCA to the 3' of the tRNA ([ec:2.7.7.25]).</p> <p>CCA-adding enzymes add the sequence [cytidine(C)-cytidine-adenosine (A)]., one nucleotide at a time, onto the 3' end of tRNA, in a template-independent reaction [[cite:PUB00081108], [cite:PUB00081109]]. This Class II group is comprised mainly of eubacterial and eukaryotic enzymes and includes Bacillus stearothermophilus CCAase [[cite:PUB00015829]], Escherichia coli poly(A) polymerase I [[cite:PUB00081110], [cite:PUB00081111]], human mitochondrial CCAase [[cite:PUB00022380], [cite:PUB00081120]], and Saccharomyces cerevisiae CCAase (CCA1) [[cite:PUB00081117], [cite:PUB00081118], [cite:PUB00081119]]. CCA-adding enzymes have a single catalytic pocket, which recognizes both ATP and CTP substrates [[cite:PUB00058171], [cite:PUB00081112]]. This family belongs to the Pol beta-like NT superfamily [[cite:PUB00013566], [cite:PUB00005745]].</p> <p>In the Pol beta-like NT superfamily [[cite:PUB00013566], [cite:PUB00005745]], the majority of enzymes have two carboxylates, Dx[D/E], together with a third more distal carboxylate, which coordinate two divalent metal cations involved in a two-metal ion mechanism of nucleotide addition. These carboxylate residues are fairly well conserved in this family.</p>
